Methotrexate may decrease folate levels in the body. Taking a high-quality folate supplement can help maintain healthy folate levels during methotrexate therapy. High-dose folate supplementation can “mask” low vitamin B12 levels, so experts recommend taking a vitamin B12 supplement along with folate.

Pyrimethamine (Daraprim): Taking folate may make this medication less effective for treating parasites.

Seizure medications (such as phenytoin [Dilantin], primidone [Mysoline], and phenobarbital [Luminal]): Folate may lower how well these medicines work and could increase the chance of seizures.
